Thromboxane A2 receptor TPβ (TPβ)

Target
TPβ
Molecular classification
G protein-coupled receptor, Receptor
01

Overview

The Thromboxane A2 receptor TPβ is a G protein-coupled receptor involved in platelet aggregation and vasoconstriction. It is one of two isoforms, TPα and TPβ, with TPβ having a longer C-terminal tail that influences receptor internalization and desensitization. TPβ is expressed in endothelial cells and plays a role in cardiovascular diseases by mediating thromboxane A2 (TXA2) effects, which are crucial in hemostasis and pathogenesis of cardiovascular disorders. Despite its potential as a therapeutic target, developing effective drugs for TPβ remains challenging due to efficacy and toxicity issues.

Mechanism of action

Activation of Gq, Gi, and Gs proteins, leading to vasoconstriction and platelet aggregation; potential drugs may inhibit these pathways to prevent thrombotic events.

Biomarkers

None specifically identified for TPβ, but TXA2 metabolites like 11-dehydrothromboxane B2 are used as indirect markers.
