Overview

Thromboxane A2 (TXA2) synthesis is a crucial pathway in hemostasis, leading to the production of TXA2, a potent eicosanoid that promotes platelet activation, aggregation, and vasoconstriction. The pathway involves the conversion of arachidonic acid to prostaglandin H2 (PGH2) by cyclooxygenase enzymes (COX-1 and COX-2), followed by the conversion of PGH2 to TXA2 by thromboxane-A synthase (TBXAS1). TXA2 acts via the thromboxane A2 receptor (TBXA2R). Inhibition of this pathway, particularly via COX inhibition by aspirin, is a key therapeutic strategy to prevent thrombotic events.
