Overview

It serves as the primary site for the assembly of the prothrombinase complex and the recruitment of additional cellular elements, which facilitates clot propagation in conditions such as deep vein thrombosis and arterial occlusion [2]. In clinical pharmacology, the thrombus surface is not a single molecular target but rather a complex biological site containing multiple therapeutic targets, including fibrin, thrombin, and various platelet adhesion receptors [3]. Thrombolytic drugs, such as tissue plasminogen activator (tPA), specifically localize to the thrombus surface by binding to fibrin, where they catalyze the formation of plasmin to dissolve the clot [4]. Additionally, targeted drug delivery systems, such as ligand-functionalized nanoparticles, are frequently designed to recognize the thrombus surface to deliver anticoagulants or thrombolytics directly to the site of vascular injury, thereby reducing the risk of systemic hemorrhage [5].
