Thymic hormones, often referred to as thymus peptides, are a group of biologically active polypeptides produced by the thymus gland that are essential for the development and regulation of the adaptive immune system [PMID: 21435018]. This group includes well-characterized molecules such as thymosin alpha-1, thymopoietin, thymulin, and thymic humoral factor [PMID: 11513737]. Their primary biological function is to promote the maturation of pro-thymocytes into functional T-lymphocytes and to modulate the activity of mature T-cells, natural killer cells, and macrophages [PMID: 17465981]. These peptides exert their effects by binding to specific receptors or pattern recognition receptors like TLR9, triggering signaling cascades that lead to the secretion of pro-inflammatory cytokines such as interferon-gamma [PMID: 25331061]. Clinically, synthetic thymus peptides like thymalfasin are used to treat chronic hepatitis B and C, as well as to boost immune responses in cancer patients undergoing chemotherapy [PMID: 18485112]. Because the term thymus peptides encompasses a variety of distinct molecules with different structures and specificities, it is considered a functional class rather than a single discrete therapeutic target.
