Overview

Thymidylate synthase (TS) is a highly conserved enzyme (EC 2.1.1.45) that catalyzes the conversion of deoxyuridine monophosphate (dUMP) to deoxythymidine monophosphate (dTMP), using 5,10-methylenetetrahydrofolate. This reaction provides the only de novo pathway for dTMP synthesis, which is essential for DNA replication and repair. Overexpression of TS has been observed in various cancers and correlates with poor prognosis and resistance to chemotherapeutic agents. It also has autoregulatory function by interacting with its own mRNA.
