Overview

Thymocyte-expressed positive selection-associated protein 1 (TESPA1) is a scaffold/adaptor protein highly expressed in developing thymocytes, peripheral T cells, and B cells. It is crucial for the positive selection and maturation of T cells in the thymus, acting as a regulator of T cell receptor (TCR) signaling. TESPA1 achieves this by binding to key signaling molecules including PLCγ1 and the inositol 1,4,5-trisphosphate receptor (IP3R), thereby facilitating TCR-triggered intracellular calcium mobilization and downstream signaling required for T cell development. It has additional roles in stabilizing TRAF6, influencing PLCγ2 phosphorylation in B cells, and modulating mast cell-mediated allergic responses. However, disruption of TESPA1 affects T cell development much more than peripheral immune responses, marking it as a stage-specific regulator within the immune system. TESPA1 is not currently targeted by approved or investigational drugs
