Overview

The Thymopoietin receptor is a cell-surface target for the thymic hormone thymopoietin and its synthetic derivative, thymopentin (TP5). It is primarily expressed on T-lymphocyte precursors and mature T-cells, where it plays a vital role in the differentiation, maturation, and functional programming of the adaptive immune system. Binding of its ligands typically triggers an increase in intracellular cyclic guanosine monophosphate (cGMP), which serves as a secondary messenger for T-cell development. While the exact molecular identity of a single "thymopoietin receptor" protein is not well-defined in modern proteomic databases—with some research suggesting interactions with the nicotinic acetylcholine receptor (nAChR) or Toll-like receptor 2 (TLR2)—it remains a recognized functional target for immunomodulatory therapy. Clinically, agonists of this receptor are used to treat immunodeficiencies, chronic infections, and as adjuvants in cancer treatment. It is frequently confused with the thrombopoietin receptor (MPL), which is a distinct protein involved in platelet production, or the thymic stromal lymphopoietin (TSLP) receptor.
