Overview

The thymoproteasome is a specialized form of the 20S proteasome complex expressed exclusively in the cortical thymic epithelial cells (cTECs). It is characterized by the presence of the unique catalytic subunit beta5t (encoded by the PSMB11 gene), which replaces the constitutive beta5 or the immuno-specific beta5i subunits found in other proteasome variants. The primary biological role of the thymoproteasome is the generation of a distinct pool of self-peptides with low affinity for MHC class I molecules, a process essential for the positive selection of CD8+ T cells and the establishment of a functional and diverse T-cell receptor repertoire. Mutations in the PSMB11 gene are linked to primary immunodeficiencies characterized by a lack of CD8+ T cells, while its unique expression profile makes it an attractive, albeit challenging, target for modulating immune responses. Although broad-spectrum proteasome inhibitors like Bortezomib interact with this complex, research is ongoing to develop subunit-specific inhibitors that could selectively modulate T-cell development for treating autoimmune disorders or enhancing anti-tumor immunity.
