Overview

Thyroid hormone receptor-interacting protein 6 (TRIP6) is an adaptor protein from the zyxin family of LIM domain-containing proteins[1][3][4][5]. It acts as a signaling platform with multidomain protein-protein interaction capabilities, localizing mainly to the cytoplasm and focal adhesions, but also shuttling to the nucleus where it functions as a cofactor in transcriptional regulation[1][3]. TRIP6 interacts with cell surface receptors (such as LPA2 and Fas/CD95), nuclear receptors (e.g., thyroid hormone receptor β, retinoid X receptor, glucocorticoid receptor), the actin cytoskeleton, and numerous kinases/adaptor proteins, thus participating in cell motility, adhesion, cytoskeletal reorganization, antiapoptotic signaling, osteoclast bone resorption, and telomere protection[1][2][3][5]. TRIP6 overexpression is implicated in cancer progression, where it regulates cell proliferation, invasion, migration, glycolysis, angiogenesis, and immune microenvironment modulation, especially in colorectal cancer[4][5]. Additionally, TRIP6 serves as both a diagnostic and prognostic biomarker in some tumor types[4]. There are no known drugs that directly target TRIP6 in clinical use, and its safety profile or direct mechanism of therapeutic action is not established.
