Overview

Thyroid hormone receptor interactor 10 (TRIP10), also known as Cdc42-interacting protein 4 (CIP4), is a member of the F-BAR protein family, which is characterized by the F-BAR (Fes/CIP4 homology-Bin/Amphiphysin/Rvs) domain important for binding and shaping cell membranes. TRIP10 is widely expressed across tissues such as kidney, brain, liver, lung, heart, and pancreas. It serves as an adaptor protein linking membrane dynamics to actin cytoskeletal reorganization, playing a key role in processes like endocytosis and vesicular trafficking. Notably, TRIP10 is crucial for the insulin-regulated translocation of the glucose transporter GLUT4 to the plasma membrane, thereby impacting glucose homeostasis. In cancer biology, TRIP10/CIP4 has been implicated in promoting tumor cell invasion and metastasis by mediating invadopodia formation. Currently, there is no evidence that TRIP10 is a direct pharmacological target, nor are there known drugs or biomarkers associated specifically with this protein. Its main scientific relevance is as an interactor in cytoskeletal remodeling and vesicle trafficking, and as a facilitator of metastatic cell behavior in some cancers.
