Overview

The thyroid hormone receptor on integrin alpha v beta 3 refers to a specific, high-affinity binding site for thyroid hormones—primarily thyroxine (T4)—located on the extracellular domain of the integrin αvβ3 heterodimer at the cell surface. This is distinct from classical nuclear thyroid hormone receptors and mediates non-genomic actions of thyroid hormones. Binding of T4 to this receptor triggers rapid activation of intracellular signaling pathways such as MAPK/ERK, leading to enhanced cell proliferation, increased angiogenesis, and modulation of apoptosis. Integrin αvβ3—and specifically its thyroid hormone receptor function—is being explored as a therapeutic target. Inhibitors like etaracizumab may be used as antiangiogenics, and other molecules such as tetrac can block or modulate this receptor's configuration/functionality related to thyroid hormone action on cancer cells.
