Overview

Thyroid hormone responsive (THRSP) is a nuclear protein that plays a key role in the regulation of lipid metabolism, specifically by promoting de novo fatty acid synthesis in tissues with active lipogenesis such as liver, adipose tissue, and lactating mammary gland[3][4]. THRSP is regulated by thyroid hormone, glucose, insulin, and nutritional factors, and its expression is notably high during periods of metabolic demand, such as lactation. In cancer, particularly breast cancer, elevated THRSP (Spot14/S14) expression correlates with poor prognosis and a more lipogenic tumor phenotype. In addition, dysregulation of THRSP may contribute to conditions such as obesity and hepatic steatosis. Although it does not act as a classical enzyme, receptor, or transporter, THRSP is considered a relevant target and biomarker in cancer and metabolic disease contexts[3][4][2][1].
