Thyroid peroxidase (TPO) is a membrane-bound enzyme essential for the synthesis of thyroid hormones. It catalyzes the oxidation of iodide and the iodination of thyroglobulin, forming T3 and T4. TPO is a major autoantigen in autoimmune thyroid diseases, and its activity is inhibited by thionamide drugs used to treat hyperthyroidism. Genetic mutations in TPO can lead to congenital hypothyroidism.
Thionamide drugs inhibit TPO activity, blocking the synthesis of new thyroid hormones.
