The thyrotropin-releasing hormone receptor (TRHR) is a G protein-coupled receptor that binds thyrotropin-releasing hormone (TRH). It plays a crucial role in the regulation of thyroid hormone homeostasis and has multiple functions in both the central nervous system and peripheral tissues. It is found on the cell membrane of thyrotropes in the anterior pituitary and triggers a series of molecular events leading to the exocytosis of thyroid-stimulating hormone (TSH) into the bloodstream.
Agonist binding to TRHR activates G proteins, leading to increased phospholipase C activity, IP3 and DAG formation, and ultimately increased intracellular calcium levels, stimulating TSH release.
