TIE-2 receptor (Tie2)

Target
Tie2
Molecular classification
Receptor tyrosine kinase (RTK)
01

Overview

TIE-2 (also known as TEK or Angiopoietin-1 receptor) is a receptor tyrosine kinase primarily expressed in endothelial cells. It plays a crucial role in blood vessel formation, remodeling, and maintenance of vascular integrity. TIE-2 is activated by angiopoietins, particularly Angiopoietin-1, which promotes vessel stability and quiescence. Angiopoietin-2 can act as either an agonist or antagonist depending on the context. Dysregulation of TIE-2 is implicated in various diseases, including vascular malformations and tumor angiogenesis, making it a potential therapeutic target.

Mechanism of action

Agonist or antagonist of TIE-2 signaling, depending on the specific agent and context. Agonists promote receptor oligomerization and autophosphorylation, while antagonists block ligand binding or downstream signaling.

Safety considerations

Potential for off-target effects due to broad expression of Tie2 in endothelial cells.Modulation of angiogenesis can have complex effects on tumor growth and metastasis.Disruption of vascular integrity may lead to bleeding or thrombosis.
