Overview

The TIE2 receptor tyrosine kinase, also known as TEK or CD202B, is a cell surface protein primarily expressed in endothelial cells. It plays a critical role in blood vessel formation (vasculogenesis and angiogenesis), maintenance, and vascular homeostasis. Ligand binding induces receptor dimerization or higher-order clustering, which is necessary for efficient signal transduction, autophosphorylation on specific tyrosine residues within its intracellular domain, and initiation of downstream signaling pathways that regulate endothelial cell behavior. Mutations in *TEK* are linked to inherited venous malformations, and TIE2 expression is associated with tumor angiogenesis.
