Overview

TIE2, also known as TEK tyrosine kinase, is a receptor tyrosine kinase predominantly expressed in endothelial cells. It plays a critical role in the regulation of angiogenesis—the formation and maintenance of blood vessels—as well as lymphatic vessel development. TIE2 is essential for vascular remodeling, maturation, and integrity during both physiological and pathological processes. Dysregulation of TIE2 signaling is implicated in several diseases, including cancer, inflammatory diseases, and congenital disorders. Pharmacological inhibition/blockade of TIE2 activity is being explored as an antiangiogenic strategy for cancer therapy.
