Tight junction protein ZO-2 (ZO-2, encoded by TJP2) is a membrane-associated scaffold protein in the tight junction multiprotein complex, crucial for assembling and regulating tight junctions in epithelial and endothelial tissues[2][3][4]. It binds transmembrane proteins and cytoskeletal elements, maintaining the paracellular barrier and cell polarity. ZO-2 is implicated in cellular signaling, gene expression, apoptosis, cell proliferation, and cytoskeletal organization. Pathogenic variants in TJP2 cause progressive familial intrahepatic cholestasis type 4, a rare but severe pediatric liver disease. Disruption of its function leads to loss of barrier integrity, altered gene regulation, and increased cell death[2][3][4].
