Tissue factor pathway inhibitor (TFPI) is a key endogenous anticoagulant protein that regulates the initiation of blood coagulation. It acts as the primary inhibitor of the tissue factor (TF)-induced coagulation pathway, thereby modulating both bleeding and clotting disorders. TFPI contains three Kunitz-type protease inhibitor domains. It inhibits early steps in blood coagulation by blocking two key procoagulant complexes, directly inhibiting FXa and inhibiting the FVIIa–tissue factor complex.
Direct inhibition of FXa via Kunitz domain 2; inhibition of FVIIa-tissue factor complex via Kunitz domain 1 upon binding to FXa.
