Overview

Tissue non-specific alkaline phosphatase (TNAP) is a crucial ectoenzyme involved primarily in skeletal mineralization through its ability to degrade inhibitors like PPi. Its activity extends beyond bones into neural tissues where it influences cell differentiation/migration. Loss-of-function mutations cause hypophosphatasia with broad clinical manifestations affecting bones/teeth/neural function. Due to its central role both physiologically and pathologically—including potential involvement in vascular calcification—TNAP remains an important target for research into metabolic bone diseases as well as extra-skeletal disorders linked to abnormal phosphate metabolism or inflammation modulation.
