The tissue regeneration environment, often referred to as the regenerative niche or microenvironment, is a complex and dynamic milieu comprising the extracellular matrix (ECM), signaling molecules such as growth factors and cytokines, and various cell types including stem cells and immune cells (Nature Reviews Molecular Cell Biology, 2017). It is not a single molecular target but rather a physiological context that orchestrates the repair and replacement of damaged tissues through coordinated signaling pathways (Science, 2012). In therapeutic contexts, researchers aim to modulate this environment to enhance healing, often by targeting specific components like transforming growth factor-beta (TGF-beta) or vascular endothelial growth factor (VEGF) to stimulate angiogenesis or reduce scarring (NIH, 2023). While critical for recovery from injury or degenerative disease, an improperly regulated regeneration environment can lead to pathological outcomes such as fibrosis or the promotion of a pro-tumorigenic niche (PubMed, 2021). Consequently, drug development typically focuses on specific receptors, enzymes, or structural proteins within this environment rather than the environment as a holistic entity.
