Tissue staining refers to laboratory techniques that use chemical dyes to color biological tissues and cells. The purpose is to increase contrast under the microscope so that cellular structures—such as nuclei, cytoplasm, organelles—or extracellular matrix components become visible and distinguishable. Stains can be classified by their origin (natural vs. synthetic), chemical properties (acidic vs. basic), solubility characteristics (water-soluble vs. lipid-soluble), mechanism of action (direct/indirect/mordant-based), and fluorescence properties. Commonly used stains include hematoxylin-eosin for general histology; methylene blue for nucleic acids; eosin Y for cytoplasmic elements; DAPI/Hoechst for DNA fluorescence imaging; among many others. Tissue staining plays an essential role in research biology as well as clinical diagnostics such as pathology and hematology.
