Tissue vasculature refers to the complex network of blood vessels, including arteries, veins, and capillaries, that permeate tissues to provide oxygen and nutrients while removing metabolic waste (NIH, 2023). It plays a fundamental role in physiological processes such as thermoregulation, immune surveillance, and the maintenance of blood pressure (StatPearls, 2023). In many pathological states, particularly cancer, the vasculature is co-opted or remodeled; for instance, tumors induce angiogenesis to sustain their rapid growth and facilitate metastasis (PubMed, 2021). Therapeutic strategies targeting the vasculature often focus on inhibiting these growth signals, such as the Vascular Endothelial Growth Factor (VEGF) pathway, or modulating vessel function to improve drug delivery. Beyond oncology, the vasculature is a primary focus in treating cardiovascular diseases, where drugs aim to manage vascular tone and prevent the formation of obstructive clots (PubMed, 2022). Because the vasculature is a systemic network, pharmacological interventions can lead to significant side effects, including hypertension, impaired wound healing, and hemorrhage (NIH, 2023).
Inhibition of vascular endothelial growth factor (VEGF) signaling, inhibition of receptor tyrosine kinases (RTKs), modulation of nitric oxide levels for vasodilation, blockade of calcium channels in vascular smooth muscle, and inhibition of coagulation factors within the vascular lumen.
