TLC domain-containing protein 1 (TLCD1) is a conserved multi-pass transmembrane acyltransferase enzyme, specifically a lysophosphatidylethanolamine acyltransferase[1]. It regulates cellular phospholipid composition by modulating the acyl chain of phosphatidylethanolamine, a key mitochondrial and plasma membrane lipid[1][2][6][7]. TLCD1 inhibits the incorporation of fluidizing phospholipids with omega-3 long-chain polyunsaturated fatty acids, thereby increasing membrane rigidity[6][7]. It is implicated in processes such as membrane assembly, phospholipid homeostasis, and membrane lipid distribution[1][2][3][5]. Disruption of TLCD1 is associated with metabolic diseases such as non-alcoholic steatohepatitis and rare genetic disorders including spastic paraplegia[1][2]. Its exact physiological and pathological roles are still being defined, but it is recognized as a molecular target in membrane biology and lipid-associated diseases[1][2][6].
