Overview

TLD domain-containing protein 2 (TLDC2) is a small eukaryotic protein primarily composed of the TLDc domain, which is structurally characterized by a globular fold with a beta-sandwich and surrounding helices[1]. TLDC2 is expressed mostly in the kidney's B-intercalated cells and some non-A non-B intercalated cells. It interacts with V-ATPase, influencing its assembly and function, thereby regulating acid-base homeostasis, particularly bicarbonate secretion during renal defense against alkalosis[2][5]. TLDC2 and its family members have a well-supported role in protection from oxidative stress, by influencing the expression and activity of antioxidant systems[1][4]. While other members of the family have been implicated in neurological disorders, TLDC2 itself has not been linked directly to human disease but is essential for proper kidney function and systemic acid-base balance[2][5].
