Overview

The TLR3–TRIF signaling pathway is a critical component of the innate immune response, primarily responsible for detecting viral double-stranded RNA (dsRNA) and initiating antiviral defenses. Toll-like receptor 3 (TLR3), upon recognizing dsRNA, activates downstream signaling exclusively through the adaptor protein TRIF (TIR-domain-containing adapter-inducing interferon-β). Upon dsRNA binding, TLR3 activates NF-κB/MAPK and IRF3/Type I Interferon pathways, leading to the production of type I interferons and proinflammatory cytokines/chemokines.
