TMC3 Antisense RNA 1 (TMC3-AS1)

Target
TMC3-AS1
Molecular classification
Long non-coding RNA (lncRNA), Antisense RNA, Intergenic lincRNA
01

Overview

TMC3 Antisense RNA 1 is a long non-coding RNA located on human chromosome 15 and is encoded antisense to the TMC3 gene. It is upregulated following inflammatory signals such as lipopolysaccharide (LPS) stimulation. Functionally, TMC3-AS1 negatively regulates the expression of interleukin 10 (IL-10) in macrophages and intestinal epithelial cells by interacting with the transcription factor p65, thereby inhibiting its binding to the IL-10 promoter. This positions TMC3-AS1 as an important regulatory element in innate immune responses. Although affiliated with inflammatory gene modulation, it does not currently serve as a classical drug target or therapeutic protein, and its disease associations require further study[1][5][7].

Mechanism of action

As a gene regulator, it may act through: Inhibition of transcription factor DNA-binding activity (e.g., blocks p65 binding at the IL-10 promoter in LPS-stimulated cells)

03

Biological functions

Regulation of inflammatory gene expression (e.g., negative regulation of interleukin 10 (IL-10))Potential transcriptional regulation of immune response genes through interactions with transcription factors (e.g., the p65 subunit of NF-κB)
04

Disease associations

Potential involvement in inflammatory diseases (regulation of innate immune response)Suspected association with Glioma Susceptibility 1 and Multiple Personality Disorder (although these links are not well-established)
05

Safety considerations

None identified; as a non-protein-coding RNA, safety concerns would pertain to theoretical RNA-based therapies
06

Biomarkers

Potential marker for inflammatory response; no validated clinical biomarker usage established
