TMEM147 antisense RNA 1 (TMEM147-AS1) is a long non-coding RNA (lncRNA) gene located on chromosome 19q13.12[3][7]. It does not code for a protein, but exerts biological effects through the regulation of other genes via RNA-mediated mechanisms. TMEM147-AS1 functions primarily as a competing endogenous RNA (ceRNA), sequestering specific microRNAs (notably miR-124 and miR-133b) and thereby modulating the expression of their target genes such as STAT3 and ZNF587[2][8]. This lncRNA has been identified as having oncogenic activity in several cancers: in estrogen receptor-positive breast cancer, its upregulation contributes to increased proliferation and invasion by altering the miR-124/STAT3 axis[2][6]; in prostatic carcinoma, TMEM147-AS1 promotes the Warburg effect (aerobic glycolysis) and tumor growth through the miR-133b/ZNF587 pathway[8]. TMEM147-AS1 is not a canonical therapeutic target such as a receptor, enzyme, or transport protein, and thus has no known approved drugs directly targeting it, nor a recognized mechanism of action for drug intervention. Its expression may have biomarker utility for cancer prognosis but is not currently used in clinical patient selection or monitoring[2][8].
