Overview

TMEM269 divergent transcript (TMEM269-DT) is a long non-coding RNA (lncRNA) locus located on chromosome 1 (chr1:42775813-42784895, hg38) and also referred to as C1orf50-AS1 or C1orf50 antisense RNA 1[1]. Like other divergent antisense lncRNAs, TMEM269-DT is transcribed in the opposite direction from a nearby or overlapping protein-coding gene, in this case TMEM269 (which encodes a transmembrane protein). Divergent transcription of lncRNA/mRNA gene pairs is a widespread feature of active gene promoters in mammals and is believed to play a regulatory role in chromatin state and gene expression, particularly in developmentally important genes and transcription factors[3][4]. These lncRNAs often function in the nucleus, associating with chromatin and potentially influencing neighboring gene expression via RNA-mediated recruitment of chromatin-modifying complexes or formation of DNA-RNA hybrid (R-loop) structures; however, for TMEM269-DT, there is currently no published evidence of a well-defined molecular or disease role, and it is not considered a therapeutic target[1][3].
