Overview

TMLHE antisense RNA 1 (TMLHE-AS1) is a long non-coding RNA (lncRNA) gene located on the X chromosome (chrX:155465970-155611616, hg38), classified specifically as an antisense lncRNA[3][9][11]. It is transcribed from the opposite strand of the TMLHE gene, which encodes trimethyllysine dioxygenase, the first enzyme in carnitine biosynthesis[6]. As an antisense RNA, TMLHE-AS1 is believed to participate in the regulation of gene expression, potentially through mechanisms like blocking mRNA translation, promoting mRNA degradation, or influencing epigenetic modifications, although its precise biological function remains experimentally unconfirmed[1][3][4][7][11]. Some indirect evidence and analogies to similar lncRNAs suggest that TMLHE-AS1 could play roles in neural cell development and differentiation in conjunction with critical transcription factors such as Sox2, but no direct interactions or therapeutic targeting have been described to date[5][7]. TMLHE-AS1 is not considered a canonical therapeutic target, receptor, enzyme, or transporter, and there are no known drugs or biomarkers associated with it[7][9][11]. Its principal importance lies in the general regulatory functions characteristic of antisense lncRNAs, rather than as a direct druggable entity.
