Overview

TNF and HNRNPL related immunoregulatory long non-coding RNA (THRIL) is a long non-coding RNA (lncRNA) that forms a ribonucleoprotein complex with heterogeneous nuclear ribonucleoprotein L (hnRNPL) and regulates transcription of the TNF-α gene by binding to its promoter[2][4]. THRIL is required for proper induction of TNF-α, a major pro-inflammatory cytokine, and modulates the expression of various immune response genes in innate immune cells such as macrophages[2][4]. Dysregulation of THRIL alters TNF-α production and is linked to pathological inflammatory responses in diseases including Kawasaki disease, tuberculosis, COVID-19, rheumatoid arthritis, and other conditions with immune and inflammatory components[1][2][4][5]. THRIL is studied as a biomarker for various inflammatory and infectious diseases, particularly for its ability to distinguish disease states in tuberculosis and its association with disease activity in Kawasaki disease and other immune-related disorders[1]. As a non-coding RNA, it is not a conventional therapeutic target (e.g., receptor or enzyme), but its central role in regulating immune gene expression makes it a subject of interest for diagnostics and possibly future RNA-based therapeutic modulation.
