Overview

TNFAIP3-interacting protein 2 (TNIP2, also known as ABIN2) is an adaptor and scaffolding protein involved in the negative regulation of NF-kappaB signaling, a central pathway in cellular inflammatory responses[1][3][5]. It contains a ubiquitin-binding UBAN domain, enabling association with linear polyubiquitin chains and coordination of protein complexes that modulate IκB kinase function, among others[1][3]. TNIP2 also stabilizes MEK kinase TPL-2 within receptor signaling complexes, impacting Toll-like receptor and MAPK pathway output[1][4][5]. Functionally, TNIP2 restrains inflammatory gene expression, mediates anti-apoptotic and anti-inflammatory signals in endothelial cells, and influences cell cycle and apoptosis dynamics in various disease models including cancer and neurodegeneration[1]. Altered TNIP2 expression links to pathologies such as cancer, inflammatory conditions, depression, osteoarthritis, trauma syndromes, and Alzheimer’s disease[1]. While no therapeutic drugs directly target TNIP2, its pivotal role in inflammatory and apoptotic signaling makes it a potential therapeutic target and biomarker for disease regulation and potential drug discovery efforts[1][5].
