Overview

TNFRSF10A Antisense RNA 1 (TNFRSF10A-AS1) is a long noncoding RNA transcribed in antisense orientation to the TNFRSF10A gene, lying in a head-to-head arrangement and possibly sharing promoter elements[2]. It is primarily localized in the cytoplasm[2][3] and participates in regulating cell death (necroptosis), inflammation, and cancer cell behaviors. Enhanced expression of TNFRSF10A-AS1 has been robustly linked to increased cell proliferation, migration, and invasion in colorectal and gastric cancer. Mechanistically, TNFRSF10A-AS1 has been shown to act as a competitive endogenous RNA, sequestering miR-3121-3p and thereby upregulating HuR, a protein that promotes tumorigenesis[3]. While evidence suggests it can regulate inflammatory and necroptotic pathways, its precise function is still being elucidated, and it is considered a promising, emerging therapeutic target in oncology[3][7].
