"Tonifying the kidney" (or Bu-Shen) is a core therapeutic principle in Traditional Chinese Medicine (TCM) rather than a specific molecular target like a receptor or enzyme. In TCM, the 'Kidney' represents a broad physiological system governing growth, development, reproduction, and bone health (Xue et al., 2022). Modern pharmacological research indicates that kidney-tonifying agents, typically complex herbal formulas or specific phytochemicals like icariin, exert their effects by modulating the hypothalamic-pituitary-adrenal (HPA) and hypothalamic-pituitary-gonadal (HPG) axes (He et al., 2017). These agents also influence key signaling pathways such as OPG/RANKL/RANK and Wnt/beta-catenin to maintain bone mineral density and inhibit osteoclastogenesis (Peng et al., 2022; Wang et al., 2023). Clinically, this approach is utilized to manage conditions such as osteoporosis, infertility, and chronic kidney disease, functioning as a multi-target therapeutic strategy rather than a single-molecule intervention (Frontiers in Pharmacology, 2022). Consequently, while it describes a recognized clinical effect, it is not classified as a molecular therapeutic target in standard biomedical databases (Zhou et al., 2022).
Modulation of the hypothalamic-pituitary-target gland axes (adrenal, gonadal, thyroid), regulation of the OPG/RANKL/RANK and Wnt/beta-catenin signaling pathways, and enhancement of antioxidant defenses to restore physiological homeostasis.
