The **tooth enamel matrix** refers to the specialized extracellular matrix secreted by ameloblast cells during tooth development which is critical for the proper formation and mineralization of enamel, the hardest tissue in the human body[3][2]. The enamel matrix is primarily composed of a unique set of proteins: **amelogenin** (the most abundant), **enamelin**, **ameloblastin**, and several proteases such as matrix metalloproteinase-20 and kallikrein-4. These proteins direct the hierarchical organization and mineralization of hydroxyapatite crystals in enamel. The matrix is transient during development—most of the protein is ultimately removed by proteolysis and reabsorption, leaving behind highly mineralized mature enamel with only trace residual protein[1][3]. Mutations in genes encoding the principal enamel matrix proteins cause various forms of *amelogenesis imperfecta*—hereditary disorders characterized by defective enamel formation[1]. In regenerative dentistry, enamel matrix derivatives (EMD), largely composed of porcine amelogenin, are used as biologic agents to stimulate periodontal tissue regeneration, where their exact mechanism remains under investigation but includes growth factor-like signaling[4].
