Topoisomerase II beta (TOP2B) is an essential enzyme that modulates the topological states of DNA during various cellular processes. It acts as a key decatenating enzyme, altering DNA topology by binding to two double-stranded DNA molecules, generating a transient double-stranded break in one molecule, passing another intact strand through this break, and then religating the broken strand. It plays a critical role in transcription regulation, chromosome segregation, DNA repair, and cell differentiation. As a therapeutic target, it is exploited by several chemotherapeutic agents, but this use also raises significant safety concerns.
Inhibits the religation step of the topoisomerase II catalytic cycle, leading to the accumulation of DNA double-strand breaks.
