Torsin family 3 member A (TOR3A) encodes a soluble AAA+ ATPase protein that resides in the lumen of the endoplasmic reticulum and the contiguous perinuclear space[2][3][11]. While the precise functions of TOR3A are poorly understood, studies suggest it may be involved in ATP-dependent protein quality control and cellular stress responses[1][3]. TOR3A is part of a family of four human Torsins (TorsinA, TorsinB, Torsin2A, Torsin3A) and shares partial functional redundancy with its family members[3]. Its activity can be regulated by cofactors, such as LULL1 (but not LAP1), which activate ATPase activity for some Torsins[3]. Genomic variants near TOR3A have been implicated in susceptibility to auto-immune ocular diseases such as Vogt-Koyanagi-Harada disease, presumably via immune and inflammatory modulation[1]. Other associations reference its induction by interferon and expression in secretory tissues, but there is no clinical evidence supporting it as a current therapeutic drug target, nor are there known pharmacological agents directed at it[2][10].
