Overview

Toxic shock syndrome toxin-1 (TSST-1) is a potent 22-kDa exotoxin and superantigen secreted by certain strains of Staphylococcus aureus [1][2]. It is the primary causative agent of toxic shock syndrome (TSS), a severe systemic illness characterized by high fever, hypotension, and multi-organ failure [2]. TSST-1 functions by binding simultaneously to the outer surface of Major Histocompatibility Complex (MHC) class II molecules on antigen-presenting cells and the variable region of the T-cell receptor (TCR) beta chain [3]. This interaction bypasses normal antigen processing, leading to the massive, non-specific activation of up to 20% of the body's T-cell population and a subsequent "cytokine storm" of pro-inflammatory mediators like TNF-alpha and IFN-gamma [1][3]. Therapeutic strategies focus on neutralizing the toxin using intravenous immunoglobulins (IVIG) or developing toxoid vaccines like STEBVax [4]. A TSST-1 toxoid is a modified, non-toxic version of the protein designed to induce protective neutralizing antibodies that block the toxin's ability to interact with immune receptors [4].
