Toxic shock syndrome toxin-1 (TSST-1) specific immunoglobulin G (IgG) and B-cell receptors (BCRs) are the primary components of the adaptive immune system responsible for neutralizing the TSST-1 superantigen produced by Staphylococcus aureus (nih.gov, 2024; Frontiers in Immunology, 2023). TSST-1 is a potent toxin that bypasses normal antigen processing by directly cross-linking MHC class II molecules with T-cell receptors, leading to a massive, non-specific activation of T cells and a life-threatening cytokine storm (mdpi.com, 2024). Specific IgG antibodies prevent this process by binding to the toxin's functional sites, thereby neutralizing its superantigenic activity and facilitating its clearance (nih.gov, 2024; mdpi.com, 2020). A lack of these protective antibodies is a major risk factor for the development of both menstrual and non-menstrual toxic shock syndrome (TSS) (Frontiers in Immunology, 2023; nih.gov, 2024). Therapeutic interventions targeting this system include the administration of intravenous immunoglobulin (IVIG) to provide immediate passive immunity and the development of recombinant vaccines, such as rTSST-1v, to stimulate long-term endogenous antibody production (nih.gov, 2024). Monitoring serum anti-TSST-1 IgG titers serves as a critical biomarker for assessing individual susceptibility to TSS and the efficacy of immunization strategies (nih.gov, 2024; ASM.org, 2024).
Neutralization of the TSST-1 superantigen by binding to its functional domains, thereby preventing the cross-linking of MHC class II molecules on antigen-presenting cells and T-cell receptors, which halts the massive release of pro-inflammatory cytokines (nih.gov, 2024; mdpi.com, 2020).
