Overview

Toxin antigens are pathogenic proteins or substances produced by various organisms—predominantly bacteria—that serve as primary virulence factors by disrupting host cellular functions and tissue integrity. These antigens are the primary targets of both active immunization through vaccines (utilizing toxoids) and passive immunization through antitoxins and monoclonal antibodies. Common therapeutically targeted toxin antigens include the Clostridioides difficile toxins A and B, the Bacillus anthracis protective antigen, and the Corynebacterium diphtheriae toxin. Drugs targeting these antigens typically act by neutralization, which prevents the toxin from binding to its specific host cell receptor or blocks its translocation and enzymatic activity within the host cytoplasm. Neutralizing these toxins is critical for preventing the severe clinical manifestations of diseases such as anthrax, tetanus, and botulism, where the toxin itself—rather than bacterial proliferation alone—drives the pathological outcome.
