Toxoplasma gondii calcium-dependent protein kinase 1 (TgCDPK1) is a vital enzyme that regulates the essential life cycle stages of the protozoan parasite Toxoplasma gondii (UniProt Q9BJF5). It acts as a primary calcium sensor, translating intracellular calcium fluxes into the activation of gliding motility, microneme secretion, and host cell invasion and egress (Lourido et al., 2010, Nature). Because TgCDPK1 is essential for parasite survival and lacks a direct ortholog in humans, it is a highly attractive therapeutic target for treating toxoplasmosis (Sugi et al., 2010, JBC). A defining structural feature of TgCDPK1 is its small glycine gatekeeper residue (G128) in the ATP-binding pocket, which is absent in nearly all human kinases. This structural difference allows for the development of selective 'Bumped Kinase Inhibitors' (BKIs) that can potently inhibit the parasite enzyme while sparing human kinases, thereby reducing potential toxicity (Ojo et al., 2010, J Med Chem).
