TPRG1 antisense RNA 1 (TPRG1-AS1)

Target
TPRG1-AS1
Molecular classification
Long non-coding RNA (lncRNA), Non-protein-coding RNA
01

Overview

TPRG1 antisense RNA 1 (TPRG1-AS1) is a human long non-coding RNA (lncRNA) that does not encode a protein, but instead functions as a regulator of gene expression, primarily via interactions with microRNAs and other gene regulatory molecules. TPRG1-AS1 is aberrantly upregulated in several malignancies, including cervical squamous cell carcinoma, where it promotes cancer cell proliferation, migration, and invasion by antagonizing the tumor-suppressive effects of miR-590-3p. It has also been shown to interact with the MYH9 protein and act as a prognostic biomarker in certain cancers. While this molecule is functionally important in disease context, it is not considered a direct therapeutic target such as a receptor, enzyme, or transporter, but instead represents a potential biomarker and mechanistic node for RNA-focused cancer research

Other names
TPRG1-AS1Tumor protein p63 regulated 1, antisense 1
02

Mechanism of action

Molecular sponge—binds to and inhibits the activity of tumor-suppressive microRNAs (such as miR-590-3p) impacting downstream gene regulation

03

Biological functions

Regulation of gene expression at the transcriptional and post-transcriptional levelNegative regulation of microRNA activity (acts as a “molecular sponge” for miR-590-3p)Modulation of cell proliferation, migration, and invasion in cancer cellsRegulation of oncogenic processes (not a receptor, enzyme, transporter, or transcription factor)
04

Disease associations

Cancer (especially cervical squamous cell carcinoma, head and neck squamous cell carcinoma, hepatocellular carcinoma, bladder cancer, colon cancer)Potential involvement in atherosclerosis
05

Safety considerations

None directly known due to non-therapeutic target status; potential off-target concerns could arise if targeted by RNA-based therapeutics, but no clinically validated inhibitors or therapies targeting this lncRNA are known
06

Interacting drugs

Propofol
07

Biomarkers

Prognostic biomarker for several cancers, notably cervical squamous cell carcinoma (association with tumor size, lymph node metastasis, and survival independently of classic pathogenic factors)
