Overview

TRAF3-interacting JNK-activating modulator (TRAF3IP3, also known as T3JAM) is an intracellular adapter protein encoded by the TRAF3IP3 gene. TRAF3IP3 regulates both innate and adaptive immunity and is essential for T and B cell development, as well as for regulatory T cell stability and function[2][8][1]. Mechanistically, it modulates signal transduction mediated by the c-Jun N-terminal kinase (JNK) pathway, interacts with TRAF3 and TBK1, and regulates the type I interferon response to cytosolic viral RNA by facilitating the degradative ubiquitination of TBK1[1][2]. It also acts in macrophages to positively regulate TLR4 signaling, contributing to inflammation, and suppresses excessive innate immune activation to maintain immune homeostasis[10][8]. TRAF3IP3 interacts with several partners, including STRN, MOBKL3, STK24, and FAM40A, and plays a role in numerous intracellular signaling pathways relevant to immune cell function and growth[3]. There are no known approved drugs that directly target TRAF3IP3.
