Overview

TRAF3-interacting protein 1 (TRAF3IP1) is a cytoplasmic protein that interacts with TNF receptor-associated factor 3 (TRAF3) and tethers it to cytoskeletal microtubules. TRAF3IP1 also acts as an inhibitor of the innate type I interferon (IFN) response and suppresses interleukin-13 (IL-13) signaling by binding to the interleukin-13 receptor alpha 1 (IL13RA1). It recruits TRAF3 and DISC1 to microtubules, playing a key role in epithelial morphogenesis, kidney development, ciliogenesis, and the regulation of microtubule cytoskeleton organization and stability via MAP4. Mutations in this gene are linked to Senior-Loken syndrome 9 and specific ciliopathies, highlighting its importance in cellular architecture and signaling[1][3][6][7][8].
