Trafficking from endoplasmic reticulum to Golgi regulator protein (TFG) is a cytoplasmic protein that plays an essential role in the dynamic function of the endoplasmic reticulum (ER), especially in the regulation of vesicle trafficking from the ER to the Golgi apparatus[3][2]. TFG is also known as the TRK-fused gene protein, having first been described as a fusion partner of NTRK1 (TRKA) in papillary thyroid carcinoma, with additional fusion partners discovered in various cancers[4][1][2]. The protein is involved in multiple processes, including protein secretion, maintenance of protein stability, autophagy, and the antiviral innate immune response, modulating NF-κB and mTORC1 signaling[1][2]. TFG’s physiological importance is underscored by its requirement for correct cargo trafficking in the secretory pathway[3][1] and by its ascribed roles in neural cell function and motor coordination, based on localization studies in brain and neuronal tissues[2]. Mutations and rearrangements involving TFG are implicated in several cancers as well as genetically defined neurologic disorders[1][2][5]. Currently, TFG is not recognized as a therapeutic target for small molecules, antibodies, or other drugs, nor are there approved biomarkers or established safety concerns related specifically to TFG as a target.
