Overview

Trafficking kinesin-binding protein 2 (TRAK2) is a cytoplasmic adaptor protein that links mitochondria and possibly other cargoes to microtubule-based motors, especially kinesin-1 and dynein. TRAK2 facilitates the bidirectional transport of mitochondria along microtubules by coordinating and activating kinesin-1 for plus-end (anterograde) movement and dynein-dynactin for minus-end (retrograde) movement. The protein plays a particularly prominent role in mitochondrial trafficking in dendrites, is involved in GABA receptor binding, and is associated with diseases such as juvenile amyotrophic lateral sclerosis. TRAK2 does not appear to be a direct therapeutic target such as a receptor or enzyme but functions as an important regulatory component in neuronal transport systems[1][2][3][4].
