Overview

Transcribed ultra-conserved region 110 (uc.110) is a member of the ultraconserved elements (UCEs), which are segments of the human genome that share 100% identity with orthologous regions in mice and rats (Calin et al., 2007, Cancer Cell). As a transcribed ultra-conserved region (T-UCR), uc.110 functions as a long non-coding RNA (lncRNA) that plays a critical role in the epigenetic and post-transcriptional regulation of gene expression. In clinical research, uc.110 has been identified as significantly overexpressed in hepatocellular carcinoma (HCC), where it acts as an oncogene by promoting cell proliferation and inhibiting programmed cell death (Braconi et al., 2011, Hepatology). Its high degree of conservation suggests it possesses essential biological functions, yet its dysregulation is a hallmark of specific malignancies. While there are currently no FDA-approved drugs targeting uc.110, experimental approaches using small interfering RNAs (siRNAs) or antisense oligonucleotides (ASOs) have demonstrated that silencing this transcript can reduce tumor cell viability. Consequently, uc.110 is being investigated as both a potential diagnostic biomarker and a novel therapeutic target for RNA-based precision medicine in oncology.
