Transcription elongation factor 1 homolog (ELOF1) is a small, evolutionarily conserved zinc-finger protein that functions primarily as an integral part of the RNA Polymerase II (Pol II) transcription elongation complex[1][3]. ELOF1 modulates transcription elongation by interacting directly with Pol II subunits and other elongation factors such as TFIIS, SPT5, SPT6, and the PAF complex[1][2][3]. Beyond its role in transcription, ELOF1 is essential for transcription-coupled nucleotide excision repair (TC-NER), promoting recruitment of repair factors (UVSSA, TFIIH) and coordinating the ubiquitylation of stalled Pol II to initiate DNA repair at transcription-blocking lesions[1][3][4]. ELOF1 deficiency leads to UV hypersensitivity, impaired transcriptional elongation rates, and increased replication stress, manifesting as genomic instability and potential synthetic lethality when combined with loss of other elongation factors[1][3]. No direct drugs or clinical biomarkers are currently associated with ELOF1, but its critical role in genome maintenance makes it a relevant molecular target for research in cancer biology and DNA repair pathways.
