Overview

Transcription elongation regulator 1 (TCERG1) is a nuclear protein that modulates transcriptional elongation and pre-mRNA splicing. It operates as a transcription factor interacting with the hyperphosphorylated C-terminal domain of RNA polymerase II via multiple FF domains, and with the pre-mRNA splicing factor SF1 via a WW domain. TCERG1 is involved in releasing RNA polymerase II from promoter-proximal pausing, thereby facilitating productive transcriptional elongation. It also plays a role in the alternative splicing of pre-mRNA and regulates the phosphorylation state of RNA polymerase II, specifically at serine 2 of its CTD. TCERG1 is necessary for TAT-dependent activation of the HIV-1 promoter and is required for efficient HIV-1 replication, making it a candidate therapeutic target in HIV-1 infection.
