Overview

Transcription elongation regulator 1 like (TCERG1L) is a protein encoded by the TCERG1L gene, a paralog of TCERG1, and functions as a regulator of transcription elongation through RNA polymerase binding and transcription coregulator activity[3][8]. TCERG1L is expressed in multiple human tissues including the brain, gut, thyroid, stem cells, tonsils, and importantly in cochlear hair cells. Genetic variants in TCERG1L (notably rs893507) are robustly associated with increased susceptibility to cisplatin-induced hearing loss in children with cancer; experimental modulation of TCERG1L expression in vitro alters cellular sensitivity to cisplatin and modulates inflammatory cytokine (IL-8, IL-6) responses, supporting its functional role in cellular stress pathways. TCERG1L has further links to inflammatory bowel disease, colon cancer predisposition, and is associated with plasma adiponectin, suggesting roles in metabolic regulation and immunity[1][2][3]. Currently, TCERG1L is not considered a classic therapeutic target like a receptor or enzyme, and there are no known drugs designed to act directly on it.
